 Electrophoretographs

Color printouts of completed sequencing electrophoretographs can be picked up in 531 Light Hall. We encourage you to examine printouts because they give an overall representation of the sequence.

 

Features

This is an example of a typical electrophoretograph. Each peak represents a different base detected by the 3730XL. Each of the four nucleotides is represented by a unique color: blue for cytosine, black for guanine, red for thymine and green for adenine. The base assignment for each peak is above the corresponding peak. Above each base assignment is the quality value. The quality value is a measure of the basecaller's accuracy. Blue bars represent high quality, yellow represent medium quality, and red are low quality base calls.


Heterozygote Basecalling

This is an example of an electrophoretograph that contains a heterozygote base. The Sequencing Analysis software allows the adjustment of a stringency threshold. The threshold is the percentage of the lower peak's height compared to the upper peak's height. In this example the threshold is set at 25%; therefore, the red peak (thymine) is at least 25% of the height of the upper blue peak (cytosine).
